Enrichment using Dynabeads or Macsbeads

Does anyone has experience using either Dynabeads or Macs beads as a negative depletion for certain subtypes before staining CyTOF samples?
I would like to enrich my PBMC popluation for CD8 cells by depleting other subtypes. Since Macs beads contain lead it might be better to use Dynalbeads? Is there somebody having experience using one or the other? It will be a negative depletion so beads will not be injected in CYTOF!

Hi,
We use the CD8 untouched Dynabeads kit. It works well with our CyTOF1.

Hi

I have used CD117 Kit enrichment from MACS. after running samples in the machine I could see some strange populations. that is probably due to bead interference. I suggest u to avoid beads in the sample.

or use negative selection method.

or u need to remove the beads from samples before running in the machine.

I have done MACS depletions and seen so issues. Couldn't find any beads that might have snuck through. Can't speak for positive selection. I have also run samples with Dynabeads in them and couldn't find bead/strange populations, so those ones are definitely safe in my book.

A member of our lab used the EasySep CD14 positive selection II to isolate monocytes that were cultured to generate macrophages. He used those macrophages as a positive control for some macrophage specific markers. He claimed that he didn't see anything funny with that data.
