Non-specific binding for Intracellular Targets

Dear Forum,

Has anyone experienced significant non-specific binding for intracellular targets? I work with different cell types such as fibroblasts in addition to immune cells and notice that there is sometimes a significant contribution from non-specific binding for intracellular targets. I have read literature where heparin was used to block non-specific binding for eosinophils. Can this treatment be used for other cell types that are not blood cells? Any feedback will be very helpful. Thank you very much!

Hi Nameera,

I would recommend that you just try staining +/- heparin to see whether it makes a difference in your fibroblasts. While the reduction in background been mostly clearly demonstrated with eosinophils, I don't think it would be completely limited to just that cell type.
