Exosome Detection

Hello!

My team and I are looking to investigate exosomes using the CyTOF and looking for some input.

Exosomes are tricky when it comes to CyTOF. It is my understanding that there are issues mainly in the detection. The reason is that often exosomes or other small extracellular vesicles cannot actually “fit” enough antibody or cell-ID reagents on the exosome to be sensed by the detector. The limit of detection is around 350 antibodies per “cell.”

We have discussed a few methods including aggregation or physically expanding the exosomes to allow for more detection signal. However, this specific work is limited.

Any methods or publications out there that might help us?

Hi Talis,
You could try using a new technology (not CyTOF) called SMCxPro by
MilliporeSigma. It is an immunoassay system (Erenna system) that can do
single molecule counting (SMC) if you have an antibody pair i.e. A capture
antibody and a detection antibody.

Here's a publication that might help you get started: they looked at synaptosomes by CyTOF.
