Maxpar kit compatability

Can the maxpar kit be used to couple metals to Fab'2 Ab fragments; or only complete Abs with intact heavy chains?

Re: Maxpar kit compatability

The MAXPAR polymers use a maleimide to covalently link to free thiols: any species with a free thiol will react with it. So,

Fab'2 structure info: http://www.abcam.com/index.html?pagecon ... &rid=12722

Therefore, if I am understanding this struture correctly, hitting a Fab'2 with TCEP to generate free thiols would make it fall apart into single Fab' units. Those could potentially be conjugated by the polymer, but I doubt this is what you want.

Also, since you've removed most of the Fc region (and its thiols), you would probably get fewer polymers/antibody, and therefore lower metal signal.
