Labeling chicken IgY

Hello everybody.

Has anyone tried labeling a chicken IgY? I am guessing it shouldn't be a problem but I was wondering if anybody has experience/ tips to share.

Looking at the structure of IgY relative to IgG, there are definitely disulfide bridges in the Fc region. Whether the lack of a real, flexible hinge region would be a problem, I don't know.


I do notice that Gallus Immunotech and most other IgY sourcing sites I've seen are generally only offering HRP or FITC-conjugated IgY, which is generally amine-chemistry (http://gallusimmunotech.com/custom-serv ... duction#i5 ). I haven't been able to find an example of maleimide/thiol chemistry on it.

Ultimately, you're just going to have to try it and see whether you get the appropriate binding and signal intensity. If it doesn't work, there appear to be a number of anti-chicken-IgY antibodies on the market, from cow and such. Since those are generally IgG, you could always do a chicken IgY primary, and come in with a cow IgG anti-chicken IgY secondary that you've labeled with MAXPAR.
