Ionpath reagents

Hi All,

Has anyone tried any of the pre-conjugated Mass Abs and/or conjugation kits from Ionpath? They have a small but sweet selection at present but some useful metals available as conjugation kits and or directly conjugated that you can't get from fluidigm (89Y/113In/115In/157Gd). It'd be great if anyone could share their experience of them and if they have tried them in either suspension/imaging studies.

Hi David,

IONpath’s pre-conjugated antibodies work well on our MIBI (although we sometimes have to use x2 higher titer to produce a similar staining intensity to that in the TDS). Same for our conjugated ones with their kits (did not compare to FDM due to FDM legal restrictions on the MIBI).

We validated all of their conjugation kits (also 89, 113, 115, 157) and they showed similar performance to FDM via suspension CyTOF.
MMI on AbC CompBeads was typically above 10E3. Biological samples were also stained similar to FDM.

Hi David,

I only tested out a couple conjugation kits (no pre-conjugated antibodies).

In my hands, the Ionpath polymer varied from "slightly better than X8" to "slightly worse than X8", with most being "basically the same as X8".

So, I think you're going to have to see how they behave in your own hands, but there's no "Wow!" difference, good or bad.

Hi Mike,

Thanks. How rarely have we had a "wow" with mass reagents in the last 5 years :cry: . A comparable kit to the X8 is ok IMO it gives fluidigm commercial competition if nothing else. We often have a need for 89Y to slot into IMC panels so an off the shelf option is attractive and fluidigm don't have this (or directly conjugated 89Y/209Bismuth for IMC) options.

You're right we have to try this and based on yours and Tomer's experience it doesn't appear we're in for nasty surprises at least!
