Hi Mike,

Thank you for clarifying spin filtration! It makes sense. One last point, and then I am out of here applying the freezing protocol also to my antibody panel:

Does adding EDTA to your AB cocktails lead to any benefit for freezing in the form of hampering AB aggregate formation? What is the rationale for adding it to your CyFACS buffer besides preventing cell clumps during the staining procedures?

Hi Daniel,

The EDTA is just part of the CyFACS buffer we use. We adopted the buffer from Evan Newell's protocol back when he was a postdoc in Mark Davis' lab at Stanford, and we've kept it ever since. We haven't noticed that it affects any staining, and it's kind of nice to have a chelator around in case there happens to be any free metal.

But a lot of people don't have any EDTA, so you should be able to leave it out if you want.
