CD16 Antibody for Fixed Cells

Hi everyone,

Hope everyone is well.

We at Tape Lab study signalling pathways within organoids. We fix our cells right from the beginning (using 4% PFA) to preserve any signalling that is occuring at that moment. We also co-culture our organoids with our cell types. This can be further read in our 2020 Nature Methods paper, Qin et al.

Saying the above, I wanted to ask if anyone has used a good CD16 clone (compatible with CyTOF of course) that also works very well with fixed cells (fixed with PFA)?

Hi Jahangir,

The B73.1 clone for CD16 is the best one that I know of for staining Fixed cells. Several vendors sell it, including Fluidigm on 165Ho (cat#3165007B).

I have only tried it on 2% PFA-fixed cells, so you're going to have to test it on your 4% PFA-fixed material.
