Do we have some way to increase the cell recovery efficiency

Hello! just saw some papers about Circulating Tumor Cells, quite few in blood, probably not possible be analyzed by CyTOF, because the cell recovery efficiency in CyTOF is low. Just wondering whether have some way to increase cell recovery efficiency in CyTOF?
As previously discussed, ''CyTOF machine itself has a cell transmission efficiency of only 20-30%. In other words, if you give it 1 million cells, the MAX you can usually expect to recover would be 200-300K cells as "Live Intact Singlets" (not "Found Cells", which includes debris and other background) . This is an inherent design issue to the CyTOF: cells stick to the sample loop, nebulizer tubing, nebulizer, and are lost to the spray chamber''.
Do we have some way to increase this cell recovery efficiency, from experiment, engineer or theory perspective?

Hi tianmo,

If you are using either the Helios PSI unit with a Helios (effectively CyTOFv3), or a Supersampler with any model of instrument, your cell recovery should be in the 40-50% range (ie, have 1M cells in your sample, should recover at least 400-500K cells). Note that a variable amount of those will still be debris, but the overall recovery is still above that of a CyTOFv1, or a CyTOFv2 using the sample loops.

In terms of working with CTCs, antigen-specific T cells, or any other super-rare cell population, you're still going to struggle with CyTOF experiments. Generally, you'll want to do some sort of enrichment to start out at a higher concentration from the beginning.
