Isolation of Human PBMCs

Hello,

I'm working on a project that involves analyzing isolated PBMCs, but am unfamiliar with Ficoll procedures. I was wondering if anyone out there has any tips about how to best isolate PBMCs for CyTOF analysis.

Thanks!

Hi Mark,

Here's a link to some HIMC protocols:

http://iti.stanford.edu/himc/protocols.html

About the 5th one down, you can see protocols for PBMC isolation from various sources.

Note: the protocol for the CPT tubes assumes you have already taken the tube and the blood, and spun them. There's a catalog # from BD for the tubes themselves, where you can get the protocol for using the tubes.

Thank you Mike. Now I'm wondering, is any one of those procedures better than the others in terms of maximizing PBMC viability?

From what I understand from our blood processing division, they're essentially the same.

The SepMate procedure is faster and easier, though, and so they have switched over to using them in most of their projects.
