Re: Ba contam in gut biopsy samples

Thanks Greg,
so, the criteria you are proposing is few hundred counts (fine) vs thousand counts in 138Ba to eliminate the sample out or just eliminate events that 138Ba > 1000? and work with low Ba+ events?
let me ask you how you evaluate/quantify spillover on M+1 and M+16. if sample is already contaminated, I won't have any control to compare in 139/153/154
without Ba. Thanks in advance

Re: Ba contam in gut biopsy samples

Yes, that's correct.

You should be able to see the M+1 or M+16 spillovers readily. Just plot 138 vs. 139 or 138 vs. 154. Since there isn't normally any background, when the spillover occurs, you'll see that you don't have any 138 high events with 0 signal on 139. This is pretty unusual, as there are typically at least a few events with 0 ion counts on any channel (you can most easily see this effect before singlet gating). I've attached an example picture of M+1 spillover from 169 into 170.

As in fluorescent flow cytometry, diagonal lines are always bad.

best of luck,

Greg
