Storage of tissue samples for CYTOF

I am doing a longitudinal study and would like to storage tissue samples (mouse liver) for a long time. I do not know how to storage the tissues. Has anyone done this? I'd be interested in people's opinions and alternative suggestions, thanks.

Re: Storage of tissue samples for CYTOF

Hi Lei Zheng,
From my practice, we store cells extracted from tissues in SVF + 10% DMSO and freez them in MrFrost box (isopentane filled walls) put in -80 °C (10.7 cells in 1ml), then in liquid nitrogen.
For thawing, put the tube in a 37°C waterbath and transfer to a plate with 9ml 37°C medium, leaving 1h at 37°C for recovery.
Wash, stain.
about 50% recovery of cells (5x10.6 cells from frozen 10x10.6 cells)
Hope it helps ;-)
Olivier
