problem in gating population using CyTOF files in FLOJO 10

Hi i am using flowjo10 software for my CyTOF 2 experimental files. but i could not able to see the population on the display, event length versus Ir193.
flowjo 10 showing 30% of the events are in the chart edges. but i tried to fix it still have problem. can anyone help me to fix the problem.

i changed to CyTOF in preferences on flowjo. but still problem in gating.

Hi,

Of the 11 FlowJo V10 revisions, and only the last 2 (10.0.7 and 10.0.7R2 the current posted) include the correct transform algorithm for the CyTOF data (ie, arcsine.) If you are using a different version, that may explain what you are seeing. Just for sanity, I pulled up my testing data in that version and it looks alright -

Hi Komal,

In FlowJo10, you have control over your axes: scaling + and -, as well as type of axis (linear, log, arcsinh, etc).

I have attached a PDF of screenshots of how to adjust your axes. I used FlowJo 10beta for this, but it should apply in the updated FlowJo10x.

To shove your points off the axis, add space to the negative (left) side of the plot by using the +/- located there (see slide 3).

Alternatively, use Cytobank, which applies the appropriate defaults automatically. I haven't had an active FlowJo license in at least 3 years.
