CyTOF spare parts in first 2 years of running?

Hi all,

We're just having our CyTOF being installed tomorrow in the core facility. We are now asked to estimate a budget we would need for *spare parts* withing the first two years of usage (while the instrument is still under warranty). So, my question is - how much hardware (roughly) have you used up/had on stock in the first 2 years of running CyTOF, beyond what was covered by the warranty? Extrapolation from a shorter period would help as well :)

For instance, a colleague made this list:

I understand you cannot possibly predict what I am going to break on the instrument :) but I would like to learn about your experiences on this.

HI Vinko,

Here at Stanford HIMC, we have 2 CyTOFv1 machines. We're running fairly constant at about $15,000/yr for spare parts in total. So, a rough estimate might be $8,000/machine/yr.

If you add in things like spin filters and blue-capped tubes to filter your sample immediately before injection, then that rises to ~$10K/machine/yr in total.


It's really difficult to put a number on that, though: the HIMC had a machine that was having arcing problems, and so we went through Rf coils and torch bodies for a while. This FY, that's fixed, but we've gone through nebulizers faster than expected due to irreparable clogs, breakage, etc.


I have attached our FY13 cost sheet as a guide.

I might suggest one other purchase that has improved CyTOF sample QC in our lab. The BioRad TC20 cell counter gives both an accurate cell count of your samples (optimizes push number/ cell recovery/ prevents overloading) and an image to access quality (cell integrity and the presence or absence of aggregates). It certainly says nothing about the quality of labeling (ie. titer) but it has limited issues with Nebulizer clogging and overloading of cells. I believe we paid around $3,000 for our unit and slides run approximately $0.50 to $0.70 per sample. -brandon
